Background. One of the most common mental diseases is depression, which is the leading cause of diseases and disability worldwide, according to the WHO. Thus, the researches on new compounds used in treating depression is very current, as well as the development of methods for their analysis. Objective of the study. To develop chromatographic methods of analysis (TLC, HPLC) in order to determine the identity, assay and specific impurities of Dioxoindolinone. Material and Methods. Chromatograph Shimadzu LC-20AD with UV-detector SPD-20A, as well as solvents and reagents based on the European Pharmacopoeia requirements were used within the study. Dioxoindolinone and possible specific impurities (initial synthetic substance, intermediate and decomposition products) were assessed. Results. The conditions of a thin layer chromatography as well as solvent and reagent systems for detection were investigated. The solvent system is optimal for separating specific impurities benzene: methanol (90:10) is optimal; the developed chromatographic plates were exposed to iodine vapor. Optimal HPLC conditions: in the mobile phase 0,2% H3PO4:ACN:H2O (1:40:60), all substance separate at different retention times and peak areas per each. Conclusion. The study has developed Chromatographic methods for the analysis of Dioxoindolinone. The above methods will enable developing of the AND of this substance. |
